Measurement of sperm acrosin is used in the investigation of male infertility, especially when the primary sperm quality factors, sperm motility, and sperm concentration are typical. In addition, according to several studies, measuring acrosin is vital for assessing sperm potential for fertilization and the successful outcome of in vitro fertilization (IVF).

Additional Information

Acrosin is an enzyme (a serine protease) found in the inner membrane of the acrosome and participates in several critical reproductive processes including, but not limited to:

  • Oocyte–spermatozoon interaction
  • Spermatozoon binding at the zona pellucida of the oocyte
  • Sperm penetration into the zona pellucida of the oocyte

There is now plenty of evidence demonstrating the importance of acrosin in the fertilization process. Infertility is generally related to lower levels of acrosin, while acrosomes are absent in sterile men. Acrosin and/or proacrosin activator inhibitors reduce entirely or obstruct the entry of spermatozoa through the zona pellucida but also the entry of spermatozoa into hamster oocytes without a zona pellucida.

Since acrosine activity is usually low in patients with low concentration or low sperm motility, testing is not necessary for these patients. In couples with unexplained infertility (with regular female tests and normal sperm basic parameters), finding typical acrosine values ​​can rule out pathological severe conditions of the spermatozoa acrosome, thus providing important information on the prognosis of infertility.
